The iPure FFPE Nucleic Acid Kits utilize silica purification technology to extract high-purity total DNA or total RNA from paraffin-embedded tissue samples. After deparaffinization of the paraffin-embedded tissues with xylene or deparaffinization solution and digestion with proteinase K, the samples are transferred to a silica membrane by adding binding solution and ethanol to adjust the binding conditions. The silica membrane specifically binds DNA & RNA while proteins and impurities are not adsorbed and removed. After two rounds of protein removal and deionization, total RNA & DNA are eluted using Elution Buffer/DEPC water.

        1.Extraction of DNA and PCR amplification from human tonsil paraffin sections:

        DNA was extracted from paraffin sections of human tonsil samples obtained from the Pathology Department of the Third Affiliated Hospital of Sun Yat-sen University. Primers targeting the KRAS gene were used to amplify fragments of 107bp and 226bp, and the HBB gene fragment of 488bp. Gel electrophoresis was performed on a 1% agarose gel for 30 minutes, as shown in Figure 1.

        2. Extraction of nucleic acid DNA from mouse testicular paraffin samples:

        DNA was extracted from paraffin samples of mouse testes obtained from the Animal Science College (Animal Pathology Research Group) of South China Agricultural University. Primers targeting the housekeeping gene GAPDH were used to amplify fragments of 218bp and 440bp, and the PCR products were subjected to gel electrophoresis. As shown in Figure 2.
