The reagent kit utilizes magnetic bead purification technology, suitable for extracting high-purity total DNA from dried blood spots, blood cards, and saliva card samples. The obtained nucleic acid can be directly used for PCR, quantitative PCR, enzyme digestion, Southern blot hybridization, and other experiments. During operation, only a 6mm diameter puncher is needed to take 1-2 pieces of blood card samples with the sample, and no centrifugation or vacuum filtration is required, suitable for use with various automated platforms.

After punching out 1 and 2 pieces with a 6mm puncher respectively, the nucleic acid was extracted using the iPure Blood Card Genomic DNA Extraction Kit (magnetic bead method), and the concentration of the extracted nucleic acid was measured using Qubit 3.0 (Thermo Fisher). The average total nucleic acid amount for 1 piece (6mm) = 58.1±SD, SD=0.114, CV=0.068 (Figure 1); the average total nucleic acid amount for 2 pieces (6mm) = 152±SD, SD=0.435, CV=0.1 (Figure 2). The reagent exhibits high repeatability and low sample variation, suitable for a wide range of applications including prenatal diagnosis, heel blood DNA extraction, STR (forensic identification), etc.
Eight pieces were punched out using a 3mm diameter puncher, with three replicates for each group. Three mainstream supplier products were compared, with 35μl elution, and the obtained DNA was detected using β-actin amplification and Qubit HS assay kit detection. The results are shown in Figure 3 and 4. Company Q (Cat NO./56304), Company R (Cat NO./11796828001), Company M (Cat NO./MD5111), the total DNA amount obtained was Company Q (Cat NO./56304) > IGE > Company R (Cat NO./11796828001) > Company M (Cat NO./MD5111).
