For cloning construction:

Customer provides plasmid as template or vector: Ensure the provided plasmid and related information match.

Plasmid: A single, bright band, dissolved in ddH2O, free from other impurities, with a total amount of approximately 1-2 μg.

PCR Products: Desired band size is correct, purified DNA has a total amount of no less than 200 ng, concentration not less than 10 ng/μl. It is recommended to provide PCR products verified by sequencing.

gDNA: After agarose gel electrophoresis, a clear main band should be visible, with a concentration ≥ 30 ng/μl, volume ≥ 20 μl, and no degradation.

cDNA: After agarose gel electrophoresis, a diffuse band ranging from 200 to 10 kb should be visible, with the central region between 1-4 Kb. Pay attention to reverse transcription quality and perform quality control.

Bacterial Liquid: Fresh bacterial liquid, ensure no contamination, avoid excessive cultivation time to prevent bacterial aging leading to plasmid extraction failure.

For site-directed mutagenesis and mutant library:
